Mouse microarrays hybridization protocols:

Several protocols are available for mouse according to the chips type used:

  • For 15k and NeuroDev2 chips (PCR products), the labelling protocol is not currently available.
  • For Synapse chips (oligonucleotides), the protocol is available in a HTML format here
  • Pour les lames 22k (oligonucléotides), the protocol is available in a HTML format here

These movies aimed at help you in the most tricky stages in DNA microarray use. All the videos on this site are in a QuickTime Movie (.mov) format. You can freely download the QuickTime visualization software from Apple.

Washing steps after pre-hybridization:

In this movie we show you a washing method using a 50ml Falcon tube. You will also see how to eliminate isopropanol excess from the slide after the last washing step after pre-hybridization.

DNA microarray hybridization:

This movie show how to prepare the hybridization chamber, to deposit the labelled cDNA target on the slide and how to cover the microarray with a coverslip.

In this movie you will see how we incubate our slides in the water bath overnight.

The first washing step :

In this movie you will see how to do the first washing step after water bath microarray incubation. This stage is very important because it's the moment where the coverslip is tearing off the slide. It is very important when the coverslip fall out at the bottom of the Falcon tube to turn inside out the slide to avoid any contact between the DNA spots and the coversplip.

DNA microarray centrifugation drying:

On this movie you will see the slide drying step. The drying technique we use rely on slide centrifugation in 96 wells microplates carrier.

DNA microarray lecture on GenePix 4000 scanner:

This movie aimed at explain how to correctly adjust the slide inside the scanner. Indeed, if the slide is not correctly adjust (label on the top), the grid will not give you the correct spot position and the result interpretation you will be wrong.
